Response Multilayered Control of T Cell Receptor Phosphorylation
نویسندگان
چکیده
2009); the catalytic activities of PTPs have been estimated to be 10to 1000-fold higher than that of tyrosine kinases (discussedby Fischer et al., 1991). Another contributing factor is the activity of inhibitory kinases such as Csk, which also constrain T cell activation (Schoenborn et al., 2009). Progress in understanding TCR triggering will require the teasing apart of these and other competing factors and the identification of those factors with the largest impact on net phosphorylation of the TCR. The effects of pervanadate on phosphorylation levels in resting T cells observed by ourselves (Figures S1B and S1C) and by others many years ago (e.g., O’Shea et al., 1992) suggest that PTPs could be key. For us, the sheer weight of the numbers also warrants their serious consideration.

O-7: Insulin Exerts a Prosurvival Effect on Human Spermatozoa via Mechanisms That Involve the Stimulation of Akt Phosphorylation
Background: The purpose of this study was to examine the impact of Insulin (INS) on human sperm function, in light of a recent proteomic analysis indicating that these cells express the INS receptor (INSR). Materials and Methods: We used Western blot and Immunocytochemical analyses. Results: Immunocytochemical analyses confirmed the presence of INSR in human spermatozoa and localized this recep...
متن کاملO-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells
Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...
متن کاملEffects of Antiproliferative Protein (APP) on Modulation of Cytosolic Protein Phosphorylation of Prostatic Carcinoma Cell Line LNCaP
Antiproliferative protein (APP) isolated from conditioned media of two androgen-independent prostatic carcinoma cell lines, PC3 and Du-145 was shown to inhibit selectively cell proliferation of androgen-dependent prostate cancer cell line LNCaP in a dose dependent manner. This protein was further purified with HPLC using hydrophobic interaction column (phenyl 5PW) and was used to study the modu...
متن کاملPhosphorylation Site Dynamics of Early T-cell Receptor Signaling
In adaptive immune responses, T-cell receptor (TCR) signaling impacts multiple cellular processes and results in T-cell differentiation, proliferation, and cytokine production. Although individual protein-protein interactions and phosphorylation events have been studied extensively, we lack a systems-level understanding of how these components cooperate to control signaling dynamics, especially...
متن کاملERK Positive Feedback Regulates a Widespread Network of Tyrosine Phosphorylation Sites across Canonical T Cell Signaling and Actin Cytoskeletal Proteins in Jurkat T Cells
Competing positive and negative signaling feedback pathways play a critical role in tuning the sensitivity of T cell receptor activation by creating an ultrasensitive, bistable switch to selectively enhance responses to foreign ligands while suppressing signals from self peptides. In response to T cell receptor agonist engagement, ERK is activated to positively regulate T cell receptor signalin...
